Tau [P301L] 是由 MAPT 基因编码的微管相关蛋白 Tau 在第 301 位密码子发生的错义突变（脯氨酸 Pro → 亮氨酸 Leu），是额颞叶痴呆伴帕金森综合征（FTDP-17）最常见且研究最深入的致病突变之一。该突变位于 Tau 蛋白的微管结合重复区（Exon 10），会显著降低 Tau 与微管的亲和力，促进其异常磷酸化、聚集形成成对螺旋细丝（PHFs）及神经纤维缠结，加速神经元退化，广泛用于构建阿尔茨海默病及 Tau 蛋白病的细胞与转基因动物模型。 |
